Opiorphin demonstrates dual-inhibitory potency on the enkephalin-inactivating ectopeptidases human NEP (hNEP) and human AP-N (hAP-N). As an analgesic, Opiorphin is 6 times stronger than the opiate morphine. Opiorphin is closely related to the rat sialorphin peptide, which is an inhibitor of pain perception and acts by potentiating endogenous micro- and δ-opioid receptor-dependent enkephalinergic pathways. In rat studies, Opiorphin suppressed pain sensation for both chemical-induced inflammation and acute physical pain. In both cases, the administered dose of 1 mg/kg provided the same painkilling power as 3-6 mg/kg of morphine. |
