JNJ-7706621是CDK和Aurora的一个双重抑制剂,在体外可以有效地抑制CDK1、CDK2、CDK3、CDK4、CDK6、Aurora A和Aurora B,对应的IC50值分别为0.009、0.004、0.003、0.058、0.253、0.175、0.011和0.015 μmol/L。JNJ-7706621选择性地抑制正常人细胞和不同类型的肿瘤细胞的增殖,JNJ-7706621对肿瘤细胞(IC50值0.112 - 0.514μmol/L)的作用是正常细胞(IC50值3.67 - 5.42μmol/L)的10倍。在人癌细胞中,不管p53、视网膜母细胞瘤或p-糖蛋白状态如何,JNJ-7706621都可以诱导细胞凋亡、抑制集落形成和细胞生长。 |
